ABSTRACT

Objective:To study performed of develop a mice model of allergic diseases induced by crude extractings from populus pollen. Methods: A total of 60 BALB/c mice were divided randomly into there groups:normal control group,Albumin Egg(OVA) group and populus pollen model group with 20 in each.Mices were repeatedly sensitized by intraperitoneal injections of OVA or crude populus pollen extract every 5 d for four doses.Five days after the last sensitization, mices were repeatedly challenged by once daily antigen from 21-25 d.The changes of inflammatory cells in bronchoalveolar lavage fluid(BALF) were stained with hematoxylin and eosin(HE) to evaluate the degree of allergic inflammation.PAS staining was used to observe the secretion of airway mucus; The changes of the nasal mucosas and lungs of mice were stained with HE to evaluate the degree of allergic inflammation. And the average optical density of IL-4 and IFN-γ positive cells in lung tissue was measured by immunohistochemistry. The total IgE in the serum was also measured by enzyme-linked immunoassay(ELISA).Results: Compared with the mice in normal control group, those in OVA group and model group developed obvious allergic inflammation in the nasal mucosas and lungs, and increased airway mucus secretion. The number of inflammatory cells including eosinophil and neutrophils markedly increased in BALF smear. The average optical density of IL-4 positive cells in lung tissue was all increased in OVA group and model group compared with those in normal control group, and the average optical density of IFN-γ in lung tissue was on the contrary. The total IgE in the serum were all increased in OVA group and model group compared with those in normal control group, and the IFN-γ in the serum was significantly reduced in OVA group and model group compared with those in normal control group. Conclusion: Taken together, crude populus pollen extract can successfully induce a mice model of allergic diseases. This model is a useful tool in studying the mechanisms of allergic disease.

ABSTRACT

OBJECTIVETo evaluate the effectiveness of locking plate external fixator in treating middle and distal tibial fractures.

METHODSFrom January 2010 to January 2013,18 patients suffered from middle and distal tibial fractures were treated by locking plate external fixator, including 11 males and 7 females, with an average age of 53.5 (ranged from 13 to 80) years old,the course of disease ranged from 2 h to 3 d. According to AO classification, 4 cases were type A,11 cases were type B and 3 were type C. Among them,6 patients were open fracture, including 2 cases with type I, 3 cases with type II and 1 case with type III, according to Gustilo classification), 12 patients were close fracture. Operation time, postoperative complications were observed, and Johner-Wruhs scoring were used to evaluate clinical outcomes.

RESULTSAll patients were followed up from 6 to 15 (meaned 11) months. Two cases occurred skin necrosis (1 case occurred bone exposure), 2 cases occurred delayed union (all were open fracture), and 1 case occurred nail infection. No screw loosening or broken occurred. According to Johner-Wruhs scoring, 10 cases obtained excellent result,6 cases good, and 2 cases fine.

CONCLUSIONLocking plate external fixator for the treatment of middle and distal tibial fractures, which has advantages of lessen damage, shorter operative time, less complications and rapid functional recovery, is one of good choice.

ABSTRACT

OBJECTIVETo develop a LC-MS/MS method for determination of astragalin in rat plasma and study its pharmacokinetics in rats.

METHODSThe analytical column was packed with ZORBAX SB-C18, and the mobile phase was methanol-water containing 10 mmol/L ammonium acetate-formic acid (80:20:0.15, V/V/V). Quercetin was used as the internal standard (IS). Multiple reaction monitoring (MRM) mode was employed, and the transition of m/z was m/z 449.1→m/z 287.1 for astragalin, and m/z 301.1→m/z 151.1 for IS.

RESULTSA good liner relationship was obtained within the range of 1.00-1000 ng/ml (r(2)=0.9929), and the lower limit of quantification of astragalin was 1.00 ng/ml in rat plasma. The extraction recoveries were all above 93%. After oral administration of astragalin, the maximum plasma concentration of astragalin was 231.1∓67.3 ng/ml and the time to reach this value was 0.5∓0.1 h, with a half-life of 3.9∓1.3 h and an AUC of 782.6∓152.8 ng·h/ml.

CONCLUSIONThe method is highly sensitive, selective and rapid for determination of the concentration of astragalin in rat plasma to facilitates the study of its pharmacokinetics.

ABSTRACT

Objective To evaluate the damage caused by high intensity ultrasound(HIU)on tumor micro- vessels. Methods Rabbit models of the VX_2 tumor were set up.The target hepatic carcinomas were treated with HIU,and the results were observed using hematoxylin-eosin(HE)staining,vascular endothelial cell biotinylated-ulex europaeus agglutininiⅠ(UEAI)immunohistochemical staining, tumor micro-vessel counts, and electron microscopy. Results Histological examination through HE staining indicated that HIU induced carcinoma vascularities with whole tumor tissue coagulative necrosis. The UEAI immunohistochemical staining of the target lesions treated by HIU was ne- gative, and complete tumor micro-vessel uhrastructure damage was observed under the electron microscope.Conclu- sion HIU can damage tumor micro-vessels thoroughly,in such way that it may inhibit tumor growth and metastasis.

ABSTRACT

The purpose of this study was to compare the occupational health concerns and opinions among 4 groups: workers, employers and managers, government officials, and health & safety managers. It could help establishing occupational health plans efficiently and providing the way to solve health problems in workplaces in the Inchon area. The delphi technique which is used for deciding group opinion was adopted for this study. Questionnaires regarding health problems and their priorities in the workplaces were sent to four groups three times. All items were measured by five degree ordinal scales. The four groups agreed with questionnaire items, improvement of working environment, occupational health concerns of the employers, the health concerns of workers, and measurement and analysis of working environment, as the upper five priorities for solving the occupational health problems. Besides with the first five priorities, health examinations, health education, and occupational diseases were suggested as important health problems in workplace.
